I got Myst ME working just fine on Windows Vista using compatibility mode, run as administrator, disable visual themes, uninstall and reinstall old Quicktime, etc. Only a few sound glitches but no freezes. Just a thought (and I know it means buying the game again) but have you tried the GOG version? (Good Old Games - just Google them), it is only a few dollars and they seem to have ironed out most of the glitches found in running Myst/Riven on newer operating systems.
Sorry to say I don't have any other answer to offer as I have never come across your problem before and given how temperemental Vista can be sometimes I'm not sure why it won't let you do what you want.
Aah, just thinking here - check the properties of the folder you are trying to save the game in - make sure it isn't 'Read Only' - can't see why it should be but worth a look.
